Isolation and Identification of Tyrosinase Inhibitor from Galla Rhois

Abstract
Two hundred-five crude drug extracts were previously screened for their tyrosinase inhibitory activity. Among them, the 50% aqueous methanols extracts of Galla Rhois showed the potent inhibitory activity (IC_(50)=0.2 §·/mL). To isolate active compounds, the 50% aqueous methanol extracts was evaporated to a small volumn in vacuo, and then fractionated by the Diaion HP-20, Sephadex LH-20 column chromatography and repeated the preparative HPLC procedure. Three major phenolic compounds were isolated, and one of them exhibiting tyrosinase inhibitory activity stronger than L-ascorbic acid (IC_(50)=70 §¶/mL) was identified as 1 2,3 4 6-penta-O-galloyl-¥â-D-glucose (IC_(50)=50 §¶/mL) by UV, IR, ©öH/^(13)C-NMR, and FAB-MS spectroscopy. This compound showed a typical noncompetitive inhibition pattern against mushroom tyrosinase.
